Ancient DNA, Dire Wolves, and De-Extinction

In the closing keynote of GEN’s “The State of CRISPR & Genome Editing” virtual summit, originally broadcast on June 11, 2025, Beth Shapiro, DPhil, Chief Scientific Officer, Colossal Biosciences and Professor of Ecology and Evolutionary Biology at University of California Santa Cruz discussed recent advances in ‘de-extinction.’
In April, Colossal announced that it had essentially resurrected the direwolf, the extinct creature made famous by George R.R. Martin, of Games of Thrones. Shapiro discusses her background and the road that led to the creation of direwolves.

In part one, Shapiro discusses her scientific journey, her trailblazing work in ancient DNA, and an update on the wooly mammoth project at Colossal with GEN’s editor in chief, John Sterling, and IPM’s North American editor, Jonathan Grinstein, PhD. In part two, the discussion with GEN editorial director, Kevin Davies, PhD, goes into much more detail about the direwolf project and the work that led up to that moment.
